Avian Influenza subtype H5N1 (AI-H5N1) is a malignant virus that very detrimental to laying chickens because it highly contagious and mutates easily. Prevention of AI-H5N1 disease in carried out by vaccination, therefore maintain the quality vaccine, continuous research needed. This study aims determine potential vaccine isolates from Bali as measured based on cellular immune response total differential leukocyte cells. Formation antibodies influenced nonspecific specific system involving leukocytes, especially lymphocytes. Total 40 layers Novogen Brown strain were used for sample, kept since age one day commercial farm Perean Village, Tabanan Regency, Bali. Laying are vaccinated at 5 weeks intramuscular injection. 20 chikens taken randomly sample. Blood draws performed once pre-vaccination five times each week after vaccination with anticoagulants. leukocytes examined an auto hematology analyzer, while leucocytes thin blood smear stained Giemsa. data analyzed means variance test followed Duncan test. Results showed could increase had significant effect mean absolute values heterophyll cells, eosinophils, lymphocytes, monocytes, but no post-vaccination basophil
